According to foreign expert, the sand bed are divided by 3 layer from top 1 inch depth the snad is no special but keep the fishes ways or bigger particle covered underneath around 2 inches to 4 inches depth, tha sand stored some germs which is reacting with oxygen and knock down the big particle size or change the solid rubbish into soluble. At last, further 2 to 3 inch sand is good for some germs which doesn't like oxygen, this is definitely necessary to change the TOXIC NO3, NO2...etc into non toxic chemical. All the captioned transfer is using diffusion method, the cycle will be non stop, but not too fast, thus, a thicker sand bed say 6 inches is good. My case, i am using 36" tank, 22 inches height, 18inches depth. Now NO2 and NO3 is keeping at a very low level, eventhough my tank is quite congested
